Tissue culture of Begonia rex plant


Begonias are usually released by conventional vegetative methods such as stem and leaf cutting. One of the drawbacks of vegetative propagation by incisions is low multiplication speed.
Plant tissue culture technology is increasingly being used for vegetative propagation of a number of begonias around the world, including Begonia rex. Planting land plant materials is very important for commercial and international production. The high quality of this substance is essential for breeders. The use of cell culture techniques for the propagation of begonia plants has been largely due to the potential of this technique to overcome many of these problems associated with conventional cultivation. Disease-free plants can be easily multiplied by new varieties and/or hybrids. Laboratory techniques are also being applied to improve the plant, such as cell fusion. Plant regeneration of somatic plants has higher genetic integration over traditional organogenesis.
Laboratory propagation by pedicel explants is based on the technique of adventurous buds. In plant plants, adventurous buds are developed from the leaves producing adventurous branches in the leaves of Begonia. The success of the clonal propagation method depends on several factors, such as genotype, media, plant growth regulator and exoplanet type, which must be tested during the process.

Begonia rex tissue culture stages


Pettiol TCRs
Culture of petyol TCLs in MS medium contains 0.2 mg in L TDZ (thidiazuron).
The onset of the stem can be observed after 3 weeks of culture.
After 8 weeks of stem onset, transfer explants to fresh MS media without any plant growth regulators for stem elongation.
Then cut the stretched branches into small discs and transfer to a fresh MS environment without any plant growth regulators to stretch most of the stem. 
After 2 weeks of elongation, transfer the drawn shoots onto the MS environment containing 1.0 mg in L BA.
TCLs Stem
Culture of thin cell stem layers in MS medium contains 0.2 mg in L BA (6-benzyladenine) and 0.2 mg in L NAA (1-naftalene acetic acid).
After 4 weeks, the production of the shoot can be seen.
Then transfer the grown shoots to a fresh MS environment containing 0.2 mg in L BA and 0.2 mg in L NAA.
Flower Shoot TCLs
Culture of flower stem cutting discs in MS media contains 0.2 mg in L TDZ (thidiazuron).
The generation of shoots can be seen after 3 weeks of culture.
Induction of root formation
When branches grow at an altitude of about 1 to 1.5 cm, with 3 to 4 leaves, they are transferred to a rooting environment.
The rooting medium contains MS medium containing 0.5 mg in L BA, 0.1 mg in L NAA, and 1 g in activated charcoal L after 45 days of cultivation.
After a few weeks, plants with developed roots and branches will be obtained.
Then transfer these bushes to the greenhouse.
acclimatization 
Once the roots are formed, transfer the seedlings to a sterile soil:sand mixture in ratio of 1:1, pH 5.8, in jam bottles for hardening.
Then, after 6 weeks, these bushes are transferred to poly bags containing the same pot mixture.
Keep plants in the greenhouse for acclimatization at 25°C and 80% humidity.
After 6 months, the plants adapt well to the environment and begin flowering.
